CD31 by Immunohistochemistry

Use
CD31 immunohistochemistry (IHC) is a stain-only technical service used to highlight endothelial-lined structures by targeting CD31, a transmembrane glycoprotein commonly present on vascular endothelial cells, megakaryocytes, histiocytes, and plasma cell subsets. It is utilized to assist in the identification and characterization of vascular neoplasms and endothelial cell involvement within histologic specimens. This service does not include diagnostic interpretation and is intended for laboratories that perform their own slide review.
Special Instructions
This test is performed as a stain-and-return (technical) service only. ARUP recommends using the Immunohistochemistry Stain Form (#32978) or electronic submission via ARUP Connect™. For clients lacking electronic ordering capability, the strain form and an ARUP client number are required. ARUP provides tissue transport kits (ARUP supply #47808) available through eSupply or Client Services to assist in safe transport; slides must be positively charged and not oven-baked. Refer to ARUP’s Immunohistochemistry Stain Offerings brochure for additional guidance.
Limitations
Specimens fixed and embedded incorrectly may yield suboptimal staining. Frozen specimens are unacceptable. Depleted specimens or those with non-representative tissue types are unacceptable. Slides must be prepared correctly (formalin-fixed paraffin-embedded cellblocks or tissue), with positively charged slides (3–5 µm thick; minimum of two slides). Precautions must be taken to avoid excessive heat and slide baking. Transport in cooled container during summer months; frozen transport is unacceptable.

Collection Instructions
Formalin fix (10 percent neutral buffered formalin) and paraffin embed specimen; cells must be prepared into a cellblock or tissue block. Transport tissue block or 5 unstained (3–5‑micron thick), positively charged slides. If sending precut slides, do not oven bake.
Storage Instructions
Room temperature or refrigerated; ship in cooled container during summer months.
Causes for Rejection
Depleted specimens; specimens submitted with non‑representative tissue type; frozen specimens.
