Use
This test is used to assess for targeted variants in solid tumor cancers, including melanoma, gastrointestinal stromal tumor (GIST), colorectal, bladder, and hepatocellular carcinomas. It aids in prognosis and treatment decisions, particularly at initial diagnosis or for refractory disease. If lung cancer is suspected, additional genetic testing for gene rearrangements or c-MET exon 14-skipping alterations may be considered. The test identifies hotspot variants in 44 cancer-related genes, contributing to personalized variant profiles for diagnosis, prognosis, and therapy response predictions.
Special Instructions
Tissue specimens must be formalin-fixed and paraffin-embedded. Cytology smears stained with Diff-Quik or Papanicolaou are also accepted. Specimens should be kept from excessive heat during transport, using a tissue transport kit. A pathology report must accompany samples. Slides will be destroyed during processing and not returned.
Limitations
This test does not detect variants outside the targeted regions or below the detection threshold. It does not identify copy number alterations, translocations, microsatellite instability, or tumor mutational burden. It is not suited to detect minimal residual disease or distinguish somatic from germline variants. Technical limitations in pseudogenes, homologous, and low mapability regions may prevent variant identification in specified exons for certain genes, as large indels over 60bp may also not be detected. Tissue samples yielding fewer than 50ng of DNA may result in suboptimal performance.

Transport block and/or slides in a tissue transport kit (ARUP supply #47808) available online through eSupply using ARUP Connect™ or contact ARUP Client Services. Resections: Transport 8 unstained 5-micron slides. (Min: 5 slides). Small Biopsies: Transport 15 unstained 5-micron slides. (Min: 10 slides).
Storage Instructions
Room temperature; Refrigerated also acceptable. Ship in a cooled container during summer.
Causes for Rejection
Less than 10 percent tumor. Specimens fixed in any fixative other than 10 percent neutral buffered formalin. Decalcified specimens. FNA smears with less than 50 tumor cells.
