Alpha-Synuclein Immunostain, Technical Component Only
Use
Alpha-synuclein is a cytoplasmic protein predominantly found in the presynaptic nerve terminal of the brain. It is involved in neuronal plasticity, synaptic function, and is abundant in Lewy bodies in Parkinson's disease and dementia with Lewy bodies. It is a major component of amyloid plaques in Alzheimer's disease, making it useful for identifying alpha-synuclein in neurodegenerative disorders.
Special Instructions
This test includes only the technical performance of the stain; no pathologist interpretation is provided. It should not be ordered with a Pathology Consultation (PATHC), as the technical component stains will be canceled if ordered concurrently. Shipping requires the use of a green 'Attention Pathology' label and a pink Immunostain Technical Only label on the transport container.
Limitations
The test does not include pathologist interpretation, only technical performance of the stain. Suitability of glass slides can affect staining results due to environmental factors that alter slide chemistry. As best practice, paraffin sections should be cut within 6 weeks to maintain immunoreactivity. Age and storage conditions of slides should be controlled to avoid compromising staining outcomes.

Submit formalin-fixed, paraffin-embedded tissue block OR 2 unstained, positively charged glass slides (25- x 75- x 1-mm) per test ordered; sections 4-microns thick.
Storage Instructions
Ambient temperature preferred for transport. Alternate temperature: Refrigerated.
Causes for Rejection
Wet/frozen tissue, cytology smears, nonformalin fixed tissue, nonparaffin embedded tissue, noncharged slides, ProbeOn slides, Snowcoat slides.
