BAP1 Immunostain, Technical Component Only
Use
BRCA1-associated protein 1 (BAP1) is involved in tumor suppression and is a member of the polycomb group proteins of transcriptional repressors. It is located on chromosome 3p21, and loss of one copy of the gene or inactivating mutations can lead to increased risk and development of tumors, including malignant mesotheliomas, uveal melanomas, clear cell renal cell carcinoma, and esophageal squamous carcinomas.
Special Instructions
This test provides only the technical staining component; no pathologist interpretation is included. When ordered with a pathology consultation, the technical components will be cancelled since interpretation is required separately. Proper completion of the Immunohistochemical (IHC)/In Situ Hybridization (ISH) Stains Request form is necessary if not ordering electronically.
Limitations
Immunohistochemistry for BAP1 expression cannot differentiate between germline and somatic mutations. The age of cut paraffin sections can affect immunoreactivity, and it's recommended that paraffin sections be cut within six weeks for best results. Environmental factors like humidity and heat can affect the charge of glass slides, potentially affecting staining quality. Improper handling might lead to diagnostic confusion.

Collection Instructions
Attach the green pathology address label and the pink Immunostain Technical Only label to the container. Collect formalin-fixed, paraffin-embedded tissue block or 2 unstained, positively charged glass slides (each 4-microns thick) per test ordered.
Storage Instructions
Minimize slide storage time after unpackaging; limit exposure to high humidity and heat; minimize exposure to plastics.
Causes for Rejection
Wet/frozen tissue, cytology smears, nonformalin fixed tissue, nonparaffin embedded tissue, noncharged slides, ProbeOn and Snowcoat slides will be rejected.
