BCOR Immunostain, Technical Component Only
Use
BCOR (BCL-6 interacting corepressor) immunohistochemistry is a highly sensitive marker in identifying round cell sarcomas with BCOR abnormalities. It is particularly useful in aiding the distinction of a subset of primitive round cell sarcomas with BCOR rearrangements from other Ewing/Ewing-like sarcomas. This test provides technical performance for detecting BCOR expression, which involves suppressing gene expression by interacting with BCL-6 or PCGF1 and inducing gene silencing by histone modification.
Special Instructions
This test includes only the technical performance of the stain with no pathologist interpretation provided. Technical component only stains should not be ordered with PATHC / Pathology Consultation, as they will be canceled if ordered together. Any immunohistochemistry (IHC)/in situ hybridization (ISH) stain performed as part of PATHC is at the discretion of the reviewing pathologist with additional charges applied.
Limitations
The age of a cut paraffin section can affect immunoreactivity. Stability thresholds for antigen detection vary widely among published literature. It is best practice for paraffin sections to be cut within 6 weeks. Environmental factors can affect the charge of glass slides, potentially altering slide staining. Unsuitable glass slides might result in inconsistent staining due to poor slide surface chemistry. Therefore, slides should be stored minimally and exposure to high humidity, heat, and plastics should be limited.
