BRG1 (SMARCA4) Immunostain, Technical Component Only
Use
This test is useful for diagnosing ovarian small cell carcinoma of hypercalcemic type (SCCOHT). Biallelic mutations in BRG1 (SMARCA4) have been reported in SCCOHT and other tumors, which exhibit loss of BRG1 protein expression in the nucleus. This loss of expression helps differentiate SCCOHT from other primary or metastatic tumors of the ovaries.
Special Instructions
This test includes only the technical performance of the stain; no pathologist interpretation is provided. If ordered with PATHC / Pathology Consultation, the technical component stains will be canceled. Any immunohistochemistry or in situ hybridization stain performed as part of the PATHC will be performed at the pathologist's discretion with an additional charge.
Limitations
The loss of BRG1 expression as determined by immunohistochemistry is not able to differentiate between germline and somatic mutations. The results should be interpreted in the context of clinical history and other diagnostic tests by a qualified pathologist. Age of a cut paraffin section and the charge of glass slides can affect immunoreactivity, leading to inconsistent staining.

Collection Instructions
Formalin-fixed, paraffin-embedded tissue block OR 2 unstained, positively charged glass slides (25- x 75- x 1-mm) per test ordered; sections 4-microns thick. Attach the green pathology address label and the pink Immunostain Technical Only label to the outside of the transport container.
Storage Instructions
Ambient (preferred). Refrigerated also acceptable.
Causes for Rejection
Wet/frozen tissue, cytology smears, nonformalin fixed tissue, nonparaffin embedded tissue, noncharged slides, ProbeOn slides, Snowcoat slides.
