CD33 Immunostain, Technical Component Only
Use
The CD33 Immunostain test is useful for the classification of myeloid neoplasms and acute leukemias. CD33 is a transmembrane protein that plays a role in cell-to-cell adhesion. It is expressed on maturing myelomonocytic cells and helps in the diagnosis of myeloid neoplasms due to its expression pattern. The presence of strong CD33 staining is indicative in classifying acute leukemias, supporting the differentiation of neoplasm types.
Special Instructions
This test includes only the technical performance of the stain, without pathologist interpretation. If a pathologist's interpretation is required, a separate order for a Pathology Consultation (PATHC) must be made. The test should not be requested with a Pathology Consultation as the technical stain component will be canceled if done.
Limitations
The test results should be interpreted by a qualified pathologist within the context of the patient's clinical history and other diagnostic tests. Certain aspects, such as age of paraffin section, can affect immunoreactivity. Environmental factors may influence the charge of glass slides, affecting the consistency of staining. It is crucial that best practices are followed in the storage and handling of slides to avoid diagnostic errors.

Collection Instructions
Formalin-fixed, paraffin-embedded tissue block, or 2 unstained, positively charged glass slides (25- x 75- x 1-mm) per test; sections 4-microns thick.
Causes for Rejection
Wet/frozen tissue, cytology smears, nonformalin fixed tissue, nonparaffin embedded tissue, noncharged slides, ProbeOn slides, Snowcoat slides.
