Claudin-1 Immunostain, Technical Component Only
Use
Claudin proteins are essential tight junction-associated proteins that prevent leakage of substances between cells. Their differential expression is observed in various epithelial and soft tissue neoplasms. Claudin-1, in particular, shows strong expression in squamous epithelial cells and may have reduced expression in some invasive breast lesions compared to benign forms. The test is useful in identifying neoplasms through claudin-1 expression patterns, being positive in about 30% to 50% of certain soft tissue and intramucosal perineuriomas, and more frequently expressed in gastric intestinal-type adenocarcinomas compared to diffuse types.

Limitations
The test does not include interpretation, only technical performance of the stain. If an interpretation is necessary, PATHC / Pathology Consultation should be ordered. Factors affecting immunoreactivity include the age of cut paraffin sections and environmental conditions affecting slide charge, leading to inconsistent staining.

Collection Instructions
Use formalin-fixed, paraffin-embedded tissue block OR 2 unstained, positively charged glass slides per test (4-micron sections). Attach specific labels to the transport container.
Causes for Rejection
Wet/frozen tissue, cytology smears, nonformalin fixed tissue, nonparaffin embedded tissue, noncharged slides, ProbeOn slides, Snowcoat slides will be rejected.
