Clusterin Immunostain, Technical Component Only
Use
Clusterin immunostaining is useful as a marker of follicular dendritic cells. In lymph nodes, tonsils, and spleen, it stains the follicular dendritic cell meshworks. B cells, T cells, and histiocytes are negative for this stain. Clusterin is often positive in tumor cells of systemic anaplastic large-cell lymphoma and usually negative in Reed Sternberg cells of classical Hodgkin lymphoma. It serves as a sensitive marker for follicular dendritic cell sarcomas.
Special Instructions
This test includes only the technical performance of the immunohistochemical stain; no pathologist interpretation is provided. Technical component only stains should not be ordered with PATHC / Pathology Consultation. If ordered with PATHC, the technical component stains will be canceled.
Limitations
The age of cut paraffin sections can affect immunoreactivity, and stability thresholds vary among antigens. Best practice is to cut paraffin sections within 6 weeks. Environmental factors can affect the charge of glass slides, impacting staining. Sending unsuitable glass slides may result in inconsistent staining due to poor slide surface chemistry. Best practices include minimizing storage time after slides are unpacked, limiting exposure to humidity and heat, and reducing contact with plastics.

Collection Instructions
Attach the green 'Attention Pathology' label and the pink Immunostain Technical Only label to the outside of the transport container. Preferred specimens include formalin-fixed, paraffin-embedded tissue block or 2 unstained, positively charged glass slides per test, sections 4-microns thick.
Causes for Rejection
Wet/frozen tissue, cytology smears, nonformalin fixed tissue, nonparaffin embedded tissue, noncharged slides, ProbeOn slides, Snowcoat slides.
