DNAJB9 Immunostain, Technical Component Only
Use
The DNAJB9 Immunostain technical component only test is useful for the diagnosis of fibrillary glomerulonephritis (FGN), a rare kidney disease. The protein DNAJB9 (Dnaj [hsp40] homolog, subfamily b, member 9) acts as a heat shock protein involved in protein folding. It is specifically identified in glomeruli of patients with FGN. By employing immunohistochemistry, an intense smudgy staining of extracellular glomerular deposits for DNAJB9 can be observed in patients suffering from this condition.
Special Instructions
This test is strictly for the technical performance of the immunohistochemical stain without pathologist interpretation. If interpretation is needed, a separate PATHC/Pathology Consultation should be ordered. The technical component stains are not to be ordered alongside PATHC, as these will be canceled if ordered together.
Limitations
The age of a cut paraffin section can affect immunoreactivity, with stability varying by antigen. Paraffin sections should ideally be cut within six weeks. Environmental factors can affect the charge of glass slides, potentially impacting slide staining quality. Storage practices for positively charged slides should minimize exposure to humidity, heat, and plastics, and the time they are stored after unpackaging should be minimized to avoid inconsistent staining.
