Integrase Interactor 1 (INI1/BAF47) Immunostain, Technical Component Only
Use
Integrase interactor 1 (INI1) is a crucial component of the SWI/SNF chromatin remodeling complex, which is involved in regulating transcription by modulating DNA accessibility for proteins. Dysfunction in these complexes has been linked to various human cancers. In pediatric rhabdoid tumors, rhabdomyosarcoma, chronic myeloid leukemia, and central nervous system tumors such as medulloblastomas and choroid plexus carcinomas, alterations in the INI1/BAF47 gene lead to loss of nuclear staining, which this test identifies. In healthy tissues, nuclear expression of INI1/BAF4 is present in all cell types.
Special Instructions
This test offers only the technical performance of the INI1/BAF47 immunohistochemical stain and does not include pathologist interpretation. The technical component stains should not be ordered with PATHC / Pathology Consultation, as they will be canceled if ordered together. Clients can opt for digital image access, but this requires electronic ordering. A manual requisition form must accompany the specimen if not ordered electronically.
Limitations
Age and environmental conditions of paraffin sections can impact immunoreactivity, potentially compromising results. Additionally, non-optimal glass slide charges can alter staining effectiveness, leading to inconsistent results. Proper storage and handling of slides and sections are essential to maintain test reliability.

Collection Instructions
Prefer formalin-fixed, paraffin-embedded tissue block, or 2 unstained, positively charged slides (25- x 75- x 1-mm) with sections 4-microns thick per test ordered.
Storage Instructions
Attach appropriate labels to transport container.
Causes for Rejection
Wet/frozen tissue, cytology smears, nonformalin fixed tissue, nonparaffin embedded tissue, noncharged or inappropriate slides.
