Mannan Binding Lectin Complement Pathway, Functional, Serum
Use
This test is useful for investigating recurrent meningococcal disease in young children and recurrent or severe infections in adults. It is also helpful for investigating glomerular kidney diseases. Additionally, deficiencies or dysregulation within the complement system may be identified in patients using this test in combination with related tests.
Special Instructions
This test is rarely useful when ordered in isolation. It should be paired with other complement pathway tests for more comprehensive evaluation. The test requires fasting preferred but not mandatory, and specimens should not be collected earlier than 48 hours following plasma exchange.
Limitations
The assay is dependent on correct sampling, storage, and shipping conditions. Degradation by temperature and consumption of complement components may lead to falsely low results. Preanalytic handling can lead to falsely low, but not falsely normal results. Functional results inconsistent with clinical history require a new blood draw. Testing should be done before plasma exchange.

Collection Instructions
Immediately after collection, place the tube on wet ice. Centrifuge at 4 degrees C after clotting on wet ice. Aliquot serum into a plastic vial and freeze within 30 minutes of centrifugation. Use dry ice if not frozen immediately.
Patient Preparation
Fasting: 8 hours preferred but not required. Do not collect earlier than 48 hours after plasma exchange.
Storage Instructions
Freeze specimen immediately after centrifugation. Specimen should remain frozen for up to 14 days.
Causes for Rejection
None based on gross hemolysis, lipemia, or icterus.
