MSH6 Immunostain, Technical Component Only
Use
This test is useful for identifying patients at high risk for hereditary nonpolyposis colorectal cancer (Lynch syndrome) and hereditary endometrial carcinoma in an immunopanel including MSH6 and other mismatch repair markers. It involves evaluating tumor tissue for mismatch repair deficiencies using immunohistochemistry (IHC) to identify patients who might benefit from further genetic testing.
Special Instructions
This test includes only the technical performance of the stain; no pathologist interpretation is provided. Technical component only stains should not be ordered with PATHC / Pathology Consultation. If ordered with PATHC, the technical components will be canceled. Attach the green 'Attention Pathology' address label (T498) and the pink Immunostain Technical Only label to the transport container.
Limitations
The test does not provide pathologist interpretation and is intended only for technical performance of the stain. Age of paraffin sections and improper slide handling can influence immunoreactivity. Stability varies among antigens. Best practice is for paraffin sections to be cut within 6 weeks and avoid high humidity and heat exposure for positively charged slides. This limit test also does not differentiate between germline and somatic mutations without genetic testing. MSI-H phenotypes and loss of protein expression in various sporadic cancers do not quickly distinguish an etiology.

Causes for Rejection
Wet/frozen tissue, cytology smears, nonformalin fixed tissue, nonparaffin embedded tissue, noncharged slides, ProbeOn slides, Snowcoat slides will be rejected.
