NUT Immunostain, Technical Component Only
Use
This test aids in the diagnosis of nuclear protein in testis midline carcinoma. It specifically detects the presence of chromosomal rearrangements involving the NUT gene on chromosome 15q14, associated with NUT midline carcinoma (NMC). NMCs are aggressive and highly lethal carcinomas that are difficult to distinguish from other poorly differentiated carcinomas using only morphological analysis.
Special Instructions
This test includes only the technical performance of the stain. No pathologist interpretation is provided. Technical component only stains should not be ordered with PATHC / Pathology Consultation. If ordered with PATHC, the technical component stains will be canceled. Clients must ensure that the correct forms are completed and attached if the test is not ordered electronically.
Limitations
The test does not include pathologist interpretation, which is necessary for a complete diagnostic evaluation. The age of the paraffin section can affect immunoreactivity, and environmental factors can impact the charge of glass slides, potentially affecting staining outcomes. Best practices for slide storage should be followed to mitigate these issues.

Collection Instructions
Preferred specimens include a formalin-fixed, paraffin-embedded tissue block or 2 unstained, positively charged glass slides (25- x 75- x 1-mm) per test ordered; sections 4-microns thick.
Storage Instructions
Ambient temperature preferred, refrigeration is acceptable.
Causes for Rejection
Rejection criteria include wet/frozen tissue, cytology smears, nonformalin fixed tissue, nonparaffin embedded tissue, noncharged slides, ProbeOn slides, and Snowcoat slides.
