PMS2 Immunostain, Technical Component Only
Use
This test is useful for identifying patients at high risk for having hereditary nonpolyposis colorectal cancer (Lynch syndrome) and hereditary endometrial carcinoma. It is performed as part of an immunopanel including PMS2 and other mismatch repair markers. This immunohistochemistry test helps to evaluate tumor tissue and identify patients at risk due to defective DNA mismatch repair.
Special Instructions
This test includes only the technical performance of the stain without pathologist interpretation. Technical component only stains should not be ordered with pathology consultation, as they will be canceled. Proper orders should include information on block numbers if sending normal and tumor blocks.
Limitations
The absence of MLH1 and PMS2 protein expression within a tumor can be associated with somatic alterations, especially in older individuals. This test does not distinguish between somatic and germline alterations, as it is not a genetic test. Additional genetic testing may be required to confirm findings and to distinguish between sporadic and hereditary cases.

Submit formalin-fixed, paraffin-embedded tissue block or 2 unstained, positively charged glass slides (25- x 75- x 1-mm) per test ordered; sections 4-microns thick.
