SDHB Immunostain, Technical Component Only
Use
This test aids in the identification of succinate dehydrogenase B (SDHB)-deficient tumors. SDHB is a part of the complex II oxidation/reduction pathway, transferring electrons from succinate to coenzyme Q. Mutations can result in paragangliomas and pheochromocytomas. It differentiates between type 1 (SDHB +) and type 2 (SDHB -) gastrointestinal stromal tumors (GIST). Endothelial cells often stain for SDHB, serving as internal controls in GIST testing.

This test is for technical performance only and does not include pathologist interpretation. Technical components should not be ordered with PATHC/Pathology Consultation. Limitations
Age and handling of specimens can affect immunoreactivity. Stability varies and is antigen dependent. Best practices include cutting paraffin sections within 6 weeks, storing labeled slides out of high humidity and heat, and minimizing exposure to plastics to preserve slide chemistry.

Preferred: Formalin-fixed, paraffin-embedded tissue block OR 2 unstained, positively charged glass slides (25- x 75- x 1-mm) sections, 4 microns thick.
Storage Instructions
Best practices for storage: Minimize time slides are stored unpackaged; limit exposure to humidity and heat.
Causes for Rejection
Wet/frozen tissue, cytology smears, nonformalin-fixed tissue, nonparaffin-embedded tissue, noncharged or inappropriate slides can be rejected.
