T-PIT Immunostain, Technical Component Only
Use
The T-PIT Immunostain is useful for the diagnosis of corticotroph, silent corticotroph, and null cell adenomas of the pituitary, which are types of pituitary tumors. These tumors can sometimes be difficult to diagnose due to their silent presentation. T-PIT, being a transcription factor of functioning and silent corticotroph adenomas, acts as a marker by which these adenomas can be identified, supporting the differential diagnosis of pituitary adenomas and differentiating null cell adenomas when T-PIT is included in a panel with transcription factors such as SF-1 and Pit-1.
Special Instructions
This test includes only the technical performance of the immunohistochemical stain, with no pathologist interpretation provided. It should not be ordered alongside PATHC / Pathology Consultation, as it will be canceled if ordered together. Attach all required labels to the shipping container as specified in the ordering instructions.
Limitations
The age of paraffin sections can significantly impact immunoreactivity. There is variability in stability thresholds across literature, making it important for cut paraffin sections to be handled properly, with cutting recommended within 6 weeks of analysis. Additionally, slide surface chemistry can be altered due to environmental exposure, affecting staining consistency. The presence of noncharged slides or incorrect slide types will cause rejection of the specimen.

Collection Instructions
Preferred: Formalin-fixed, paraffin-embedded tissue block OR 2 unstained, positively charged glass slides (25- x 75- x 1-mm) per test ordered; sections 4-microns thick.
Storage Instructions
Attach appropriate labels to the outside of the transport container.
Causes for Rejection
Wet/frozen tissue, cytology smears, nonformalin fixed tissue, nonparaffin embedded tissue, noncharged slides, ProbeOn slides, Snowcoat slides.
