BOB.1, technical only
Use
BOB.1 is a marker used to identify B-cells in germinal centers, mantle cells, and lymphomas. It helps determine the lineage of CD20 negative B-cell neoplasms and differentiate between classical Hodgkin's lymphoma, which is BOB1 negative, and primary mediastinal large B-cell lymphoma, which is positive. This test is technical only, meaning it does not include a pathologist read.
Special Instructions
Fresh tissues should be fixed at a 20:1 fixative to tissue ratio, in 10% neutral buffered formalin (NBF) for 6-72 hours, with 48-72 hours preferred. Formalin-fixed paraffin embedded tissue (FFPE blocks) should be formalin fixed. Unstained slides should be 2 slides per stain or a minimum of 1 slide per stain, and be formalin fixed. Slides should be left unbaked and positively charged to prevent background staining and tissue loss.
Limitations
Specimens should not be fixed in any fixative other than 10% NBF or B plus, as all other fixatives will be rejected. Fresh tissues should be tested within 72 hours if fixed in formalin. Samples transferred to 70% ethanol after fixation should be tested within 3 months for accurate results.
