Use
CD25 is the IL-2 receptor alpha chain (IL2RA) present on activated T-cells. The receptor is known to be upregulated in conditions like EBV-transformed B-cells, Hairy cell leukemia, and systemic mastocytosis. Clinically, CD25 expression is significant as it is also observed in inflammatory and malignant conditions that increase IL2 uptake, such as lymphoid neoplasms, autoimmune disorders, and allograft rejection. This provides insight into disease activity and potential therapeutic targets.
Special Instructions
Fresh tissues may be stored in formalin for 24-72 hours before being transferred to 70% ethanol for long-term storage at 2-8°C. Unstained slides must be unchecked with positively charged glass slides to prevent background staining and tissue loss.
Limitations
Not all conditions associated with CD25 expression are detected by this test alone; variable staining might be seen in macrophages, and other tests may be required for comprehensive analysis of certain diseases or conditions.

Collection Instructions
Fresh tissues should be fixed in a fixative to tissue ratio of 20:1 with 10% neutral buffered formalin for 6-72 hours, preferably 48-72 hours.
Storage Instructions
Temperature 15°C to 25°C for storage.
Causes for Rejection
Specimens not fixed in 10% NBF or B plus will be rejected.
