Use
CD25 is the IL-2 receptor alpha chain (IL2RA) found on activated T-cells. This receptor is known to be upregulated in conditions like EBV-transformed B-cells, Hairy cell leukemia, and systemic mastocytosis. It's often found in cases where inflammatory and malignant conditions increase IL2 uptake, such as lymphoid neoplasms, autoimmune disorders, and allograft rejection. Variable staining may be seen in macrophages. It is important to note this test is only for the technical component, not including pathologist review.
Special Instructions
Fresh tissues should be fixed with a fixative to tissue ratio of 20:1 using 10% neutral buffered formalin (NBF) for 6-72 hours, with a preference for 48-72 hours. Specimens may later be transferred to 70% ethanol and stored at 2-8°C for longer-term storage if needed. Positively charged glass slides are required for unstained slides to prevent background staining and tissue loss, and should be left unbaked.
Limitations
This test does not include a pathologist read, meaning the technical execution is covered without interpretation. Results can be affected by preservation methods and timing; only specimens fixed in 10% NBF or B plus will be accepted. Other fixatives will lead to rejection. Variable CD25 staining may impair consistent results.
