Use
The CD3 antigen is a marker of T-cell differentiation, detectable from early thymocytes to mature T-cells. It is used in conjunction with other antibodies to characterize T-cell disorders. Staining variability may be observed on natural killer cells.
Special Instructions
Fresh tissues must be fixed in neutral buffered formalin (10% NBF) for 6-72 hours with a preferred duration of 48-72 hours. Specimens can be further stored in 70% ethanol at 2-8°C for long-term storage. Unstained slides should be on positively charged glass slides to prevent background staining and tissue loss.
Limitations
Variable staining can occur in natural killer cells, which might affect interpretation. All specimens must be appropriately fixed in 10% NBF, as alternate fixatives will lead to specimen rejection.
