CD45, technical only
Use
CD45 (leukocyte common antigen) is a glycoprotein ubiquitously expressed on the cell surface of nucleated hematopoietic cells (including lymphocytes, monocytes, and eosinophils), but is absent from erythrocytes and platelets. Expression of CD45 is necessary in the regulation and activation of the immune system. The test is technical only and does not include a pathologist read.
Special Instructions
CD45 testing is technical only, requiring submission of correctly prepared tissues or slides as per specimen requirements. Specimens must be fixed in 10% neutral buffered formalin or equivalent to avoid rejection. For fresh tissues, the use of a 70% ethanol solution is recommended for transportation after fixation. Positively charged glass slides are required for unstained slides to prevent background staining and tissue loss.
Limitations
The test does not include a pathologist interpretation. CD45 expression may vary based on cell type, state of differentiation, or activation status. Unstained slides must be unbaked to avoid interference with staining quality. Fixation conditions play a critical role and must be adhered to for accurate results.

Collection Instructions
Fixative should be at a 20:1 fixative to tissue ratio, 10% neutral buffered formalin (NBF) for 6-72 hours (48-72 hours PREFERRED).
Storage Instructions
15°C to 25°C. Use a refrigerated (NOT FROZEN) gel pack in the shipment to protect from extreme temperatures; separate gel pack from specimen.
Causes for Rejection
Specimens must be fixed in 10% NBF or B plus. All other fixatives will be rejected.
