Congo Red Stain, technical only
Use
Congo red special stain is used to detect amyloid deposits in tissue sections. Congo red binds to amyloid fibrils, and under polarized light, these deposits exhibit characteristic apple-green birefringence, aiding in the diagnosis of amyloidosis. This staining is essential for identifying amyloid-related diseases, as it highlights the presence of amyloid deposits that may not be visible with routine histological stains.

Limitations
Specimens must be fixed in 10% neutral buffered formalin (NBF) or B plus. All other fixatives will be rejected. Congo Red stain is intended for the detection of amyloid deposits and is not inclusive of pathological evaluation. It should be noted that staining and artifact may vary depending on the fixation method and tissue preparation.

Collection Instructions
Fixative should be at a 20:1 fixative to tissue ratio, 10% neutral buffered formalin (NBF) for 6-72 hours (48-72 hours PREFERRED). Specimens may be transferred to 70% alcohol for transportation purposes after fixation.
Storage Instructions
Transfer to ethanol for long-term storage if necessary. Keep at 2-8°C.
Causes for Rejection
Specimens fixed in anything other than 10% NBF or B plus.
