Cyclin D1/PRAD1, technical only
Use
Cyclin D1 is a cell cycle regulator that controls the transition from G1 to S phase in normal tissues. A mutation in this protein leads to increased cell proliferation, disrupting downstream pathways, and resulting in tumorigenesis. Immunohistochemistry detection is valuable in diagnosing and prognosing various tumors, including mantle cell lymphoma.
Special Instructions
Technical component only. Pathologist read not included. Fresh tissues must be fixed at a 20:1 fixative to tissue ratio with 10% neutral buffered formalin for 6-72 hours (48-72 hours preferred). Formalin-fixed paraffin embedded tissue blocks and unstained slides should be stored at ambient temperature with a refrigerated gel pack for protection. Specimens may be transferred to 70% alcohol after fixation for transportation purposes.
Limitations
Specimens must be fixed in 10% NBF or B plus; other fixatives are rejected. Fresh tissues must be tested within 3 months after placing in ethanol for storage. Positively charged glass slides are required to prevent background staining and tissue loss. Unstained slides should not be baked.
