Use
Cyclin-dependent kinase-4 (CDK4) is a cyclin-dependent kinase that associates with D-type cyclins. It plays a significant role in the progression of cells from G1-phase to S-phase of the cell cycle. This antibody does not cross-react with other members of the CDK family. CDK4 may be used to differentiate benign tumors from sarcomas and osteosarcomas. This test includes only the technical component; a pathologist read is not included.
Special Instructions
The test is technical only, meaning it does not include a pathologist's read. Fresh tissues should be fixed in formalin for 24-72 hours before transferring to 70% ethanol for longer-term storage at 2-8°C. Unstained slides require positively charged glass slides to avoid background staining and tissue loss, and should be left unbaked.
Limitations
The antibody is specific to CDK4 and does not cross-react with other CDK family members. The test is limited to technical analysis only and does not include interpretive pathology. Specimens must be fixed in 10% NBF or B plus, as other fixatives will result in rejection. Specimen stability varies, with fresh tissues stable for up to 72 hours, while FFPE and slides have indefinite stability.

Collection Instructions
Fixative should be 20:1 fixative to tissue ratio using 10% neutral buffered formalin (NBF) for 6-72 hours, preferably 48-72 hours. Specimens may be transferred to 70% alcohol for transportation after fixation.
Storage Instructions
Store at ambient temperature. For longer-term storage, specimens fixed in formalin may be transferred to 70% ethanol and stored at 2-8°C.
Causes for Rejection
Specimens not fixed in 10% NBF or B plus will be rejected.
