Epithelial membrane antigen
Use
EMA is a transmembrane protein normally expressed in glandular or luminal epithelial cells and hematopoietic cells and absent in skin epithelium and mesenchymal cells. Overexpression is common in malignancies and leads to tumorigenesis. EMA is highly expressed by most carcinomas (especially adenocarcinomas) and hematologic cancers.
Special Instructions
Fresh tissues should be fixed in formalin for 24-72 hours by collection sites, then transferred to 70% ethanol for long-term storage if needed. Positively charged glass slides are required to prevent background staining and tissue loss in unstained slides. Slides should be left unbaked.
Limitations
The accuracy of the test results may be affected by factors such as improper specimen collection, inadequate fixation, or improper storage conditions. Specimen fixatives other than 10% neutral buffered formalin or B plus will lead to specimen rejection. Interpretation of results should consider the specific clinical context of each patient.

Collection Instructions
Fixative should be at a 20:1 fixative to tissue ratio, 10% neutral buffered formalin (NBF) for 6-72 hours (48-72 hours PREFERRED).
Storage Instructions
Store at 15°C to 25°C; specimens may be transferred to 70% alcohol for transportation purposes after fixation.
Causes for Rejection
Specimens must be fixed in 10% NBF or B plus. All other fixatives will be rejected.
