Granzyme B, technical only
Use
Granzyme B is a neutral serine protease stored in specialized lytic granules of cytotoxic T lymphocytes (CTL) and natural killer (NK) cells. Granzyme B induces DNA fragmentation and apoptosis in target cells when released by CTL and NK cells. Technical component only. This test does not include a pathologist's interpretation.
Special Instructions
Fresh tissues should be fixed at a 20:1 fixative to tissue ratio with 10% neutral buffered formalin (NBF) for 6-72 hours, preferably 48-72 hours. Unstained slides must be positively charged to prevent background staining and loss of tissue.
Limitations
This test is for the technical component only and does not include a pathologist's interpretation. The fixation of specimens in formalin is mandatory, and noncompliance with this requirement will result in rejection of samples. Continuous exposure to formalin beyond recommended times or exposure to incorrect storage conditions may compromise tissue integrity. Turnaround time may vary based on clinical requirements and protocol specifications for biopharma studies.
