Use
The Myogenin test is used to detect myogenic transcriptional regulatory protein expression, which occurs early in skeletal muscle differentiation. It is also expressed in rhabdomyosarcoma, a type of soft tissue sarcoma, making this test valuable in the diagnostic process of muscle-related tumors.
Special Instructions
This is a technical component only test, which means the pathologist's interpretation is not included. Fresh tissue specimens can be fixed in formalin for 24-72 hours, then transferred to 70% ethanol for longer-term storage. Positively charged slides are required to prevent background staining and tissue loss. Slides should be left unbaked.
Limitations
The test does not include a pathologist's interpretation. The expression of myogenin may be seen in several types of soft tissue tumors, which may result in diagnostic overlap. Specimens not fixed in the specified 10% neutral buffered formalin (NBF) may be rejected, limiting test availability.

Collection Instructions
Fixative should be at a 20:1 fixative to tissue ratio using 10% neutral buffered formalin (NBF) for 6-72 hours, with 48-72 hours being preferred. Specimens must be tested within 3 months after placing in ethanol.
Storage Instructions
Store at 15°C to 25°C. Ambient shipping recommended. Use a refrigerated gel pack to protect from extreme temperatures.
Causes for Rejection
Specimens not fixed in 10% NBF or B plus. All other fixatives will be rejected.
