MGMT Promoter Methylation Analysis
Also known as: MGMT Methylation
Use
The test uses bisulfite modification of tumor DNA and real-time PCR to quantify CpG methylation within the MGMT gene promoter. It is a qualitative analysis that serves to assess the methylation status of the MGMT gene promoter which has implications in the management of certain cancers.
Special Instructions
As of May 12, 2025, this test replaces the Brain NGS Fusion Panel and other tests that are no longer offered. Paraffin block is preferred for specimen requirements.
Limitations
Detection of MGMT promoter methylation analysis is qualitative, which may limit quantitative assessments.

Collection Instructions
Paraffin block is preferred. Alternatively, send 1 H&E slide plus 5-10 unstained slides cut at 5 or more microns. Use positively-charged slides and 10% NBF fixative. Do not use zinc fixatives. A minimum of 40% tumor is required.
Storage Instructions
Use cold pack for transport, ensuring it is not in direct contact with the specimen.
Causes for Rejection
If the tumor cannot be enriched to at least 40%, then testing will not be performed.
