NKX2.2 Immunohistochemistry (IHC)
Use
Homeobox protein NKX2.2 plays a critical role in neuroendocrine/glial differentiation. The NKX2.2 gene was recently identified as a target of EWS-FLI-1, the fusion protein specific to Ewing sarcoma. NKX2.2 is a valuable marker for Ewing sarcoma with a sensitivity of 93% and a specificity of 89%, and aids in the differential diagnosis of small round cell tumors.
Special Instructions
Block and slide identifiers should be clearly written and match exactly with the specimen ID and specimen labeling as noted on the requisition. Use cold pack for transport, avoiding direct contact with specimen. NYS clients please provide date and time of collection.
Limitations
The test provides results based on immunohistochemistry techniques and may not detect all variations of NKX2.2 expression or mutations. Pathological conditions other than Ewing sarcoma can sometimes express this protein artifactually.

Formalin-fixed, paraffin-embedded (FFPE) tissue block preferred or 1 unbaked, unstained H&E slide and 2-3 positively charged unstained slides, cut at 4-5 microns.
