USP6 Rearrangement

Use
Detection of rearrangements involving the USP6 gene (17p13.2) can aid in the diagnosis of nodular fasciitis and aneurysmal bone cysts, which often harbor USP6 gene rearrangements as a neoplastic driver. This FISH-based assay provides structural rearrangement analysis useful in distinguishing these entities from histologic mimics when used in conjunction with pathologic evaluation.
Special Instructions
Level of Service: Global or Tech Only. For Tech Only service, send circled H&E slide. Reminder that decalcified specimens may be incompatible and may limit testing. Use cold pack for transport; do not place cold pack in direct contact with specimen. New York clients must provide date and time of collection.
Limitations
This FISH assay detects USP6 rearrangements but does not identify the translocation partner. Decalcified specimens may fail due to incompatibility with FISH. Limited acid decalcification may further reduce assay performance.

Collection Instructions
Send paraffin block. Also send circled H&E slide for tech‑only (required). Cut slides: H&E slide (required) plus 4 unstained slides cut at 4‑5 microns. Circle H&E slide for tech‑only. Reminder about decalcified specimens: Testing will be attempted but most decalcification solutions are incompatible with FISH. Limited acid decalcification.
Storage Instructions
Use cold pack for transport, making sure cold pack is not in direct contact with specimen.
