Cortisol, LC/MS, Saliva
Also known as: Compound F, Hydrocortisone
Use
Measurement of cortisol concentration in saliva via liquid chromatography–tandem mass spectrometry (LC‑MS/MS) provides a noninvasive method to assess endogenous cortisol levels. Salivary cortisol reflects free cortisol and is useful in evaluating adrenal function and disorders of the hypothalamic‑pituitary‑adrenal axis.
Special Instructions
Saliva collection should be performed using a Salivette® Cortisol (blue screw‑cap). Preferred volume is 0.5 mL (minimum 0.2 mL). Alternate acceptable kits: Super SAL™ or Super SAL2™ Universal Saliva Kit. Collection must occur at least 60 minutes after brushing teeth, eating, oral medication, and 10 minutes after rinsing mouth. The swab is placed in the cheek for ~2 minutes; refrigerate immediately. White‑top Salivette® devices are rejected.
Limitations
Hemolyzed specimens and use of white‑top Salivette® collection devices are grounds for rejection. Contamination by food, drink, dental products, or medications may interfere with cortisol measurement. Limited volume may necessitate repeat collection.
