Use
BRAF V600E is a protein marker targeting a mutation in the BRAF gene, substituting valine with glutamic acid at position 600. This mutation activates the MAPK signaling pathway and promotes tumorigenesis. It is frequently used as a diagnostic and prognostic marker for cancers such as melanoma, colorectal cancer, Hairy cell leukemia, and thyroid cancer.

Limitations
Specimens must be fixed in 10% neutral buffered formalin (NBF) or B plus. All other fixatives will be rejected. Slides should not be baked to prevent background staining.

Collection Instructions
Fixative should be at a 20:1 fixative to tissue ratio, 10% neutral buffered formalin for 6-72 hours (48-72 hours preferred).
Storage Instructions
Store at 15°C to 25°C. Specimens may be transferred to 70% alcohol for transport after fixation.
Causes for Rejection
Specimens not fixed in 10% NBF or B plus.
