The PD-L1 IHC (Nivolumab) test is designed to evaluate the expression of the PD-L1 protein (Programmed Death Ligand 1) using immunohistochemistry in patients with squamous cell carcinoma of the head and neck. Positive PD-L1 expression, which is indicated by the presence of membrane staining in tumor cells, is relevant for determining patient eligibility for treatment with nivolumab, an immune checkpoint inhibitor (OPDIVO). This inhibitor works by disrupting PD-1 mediated signaling to restore antitumor immunity, offering a novel therapeutic approach to extend survival and potentially improve the quality of life in patients with advanced squamous cell carcinoma of the head and neck.

Limitations

The test may not be suitable for tissue samples that have been exposed to equivalent fixation methods other than the prescribed 10% neutral buffered formalin. There is a potential for variability in PD-L1 expression results due to differences in testing protocols or tissue handling. Additionally, the test is only applicable for squamous cell carcinoma of the head and neck and is not intended for use with other tumor types. Samples insufficient in quantity or not properly preserved may be rejected.
