Periodic Acid-Schiff Stain, technical only
Use
Periodic Acid-Schiff (PAS) stain detects polysaccharides like glycogen and mucosubstances including glycoproteins and mucins in tissues. It helps identify pathological conditions such as glycogen storage diseases, adenocarcinomas, fungal infections, alpha-1 antitrypsin deficiency, mycosis fungoides, and Sézary syndrome. This test is technical only, without a pathologist's interpretation.
Special Instructions
The test is technical only and does not include a pathologist read. The unstained slides required must be on positively charged glass slides to prevent background staining and tissue loss. Samples should be tested three months after placing in ethanol storage if needed.
Limitations
The test does not include a pathologist read and is for technical performance only. Samples may degrade beyond the recommended storage stability, affecting results. Only specific fixatives are accepted to ensure test validity, and any deviation may result in specimen rejection or compromised results.

Collection Instructions
Fixative should be at a 20:1 fixative to tissue ratio, 10% neutral buffered formalin (NBF) for 6-72 hours (48-72 hours PREFERRED).
Storage Instructions
Store at 15°C to 25°C. Samples may be transferred to 70% ethanol for longer storage and tested within three months.
Causes for Rejection
Specimens must be fixed in 10% NBF or B plus. All other fixatives will be rejected.
