Periodic Acid-Schiff Stain with Diastase, technical only
Use
The PAS-D (Periodic Acid-Schiff with Diastase) special stain is used in pathology to identify glycogen, mucopolysaccharides, and certain fungal organisms in tissue samples. Diastase treatment specifically breaks down glycogen, helping to differentiate it from other structures. This stain is commonly used in diagnosing glycogen storage diseases, fungal infections, and various disorders involving mucopolysaccharides. Technical component only. Pathologist read not included.
Special Instructions
Fresh Tissue specimens may be fixed in formalin for 6-72 hours by collection sites, then transferred to 70% ethanol and stored at 2-8°C for longer term storage if needed. This is NOT preferred. Samples should be tested 3 months after placing in ethanol. Unstained slides require positively charged glass slides to prevent background staining and tissue loss. Slides should be left unbaked.
Limitations
This test is for technical execution only and does not include pathologist interpretation or final diagnostic reporting. Various factors such as inadequate fixation, improper shipping, and inappropriate storage conditions can affect the quality and interpretability of the staining result.

Collection Instructions
Fixative should be at a 20:1 fixative to tissue ratio, 10% neutral buffered formalin (NBF) for 6-72 hours (48-72 hours PREFERRED)

Causes for Rejection
Specimens must be fixed in 10% NBF or B plus. All other fixatives will be rejected.
