Tyrosinase (TYROS) Immunostain, Technical Component Only
Use
Tyrosinase is expressed in the majority of melanomas, making it a useful diagnostic marker. This antibody will detect the tyrosinase enzyme in the cytoplasm of normal melanocytes and cells of malignant melanoma. It is used to aid in the identification of normal melanocytes and malignant melanoma.
Special Instructions
This test includes only the technical performance of the stain and does not provide pathologist interpretation. Technical component only stains should not be ordered with PATHC / Pathology Consultation. If ordered with PATHC, the technical component stains will be canceled. The shipment requires specific labeling: attach the green 'Attention Pathology' label and the pink Immunostain Technical Only label to the transport container.
Limitations
This test does not include pathologist interpretation, only the technical performance of the stain. Best practice dictates that paraffin sections be cut within 6 weeks, as the age of a cut section can affect immunoreactivity. The charge of glass slides may affect slide staining due to environmental factors, resulting in inconsistent staining.

Collection Instructions
Preferred specimens: Formalin-fixed, paraffin-embedded tissue block or 2 unstained, positively charged glass slides (25- x 75- x 1-mm) per test ordered, sections 4-microns thick.
Storage Instructions
Best practices for storage of positively charged slides: minimize time slides are stored after being unpackaged, limit exposure to high humidity and heat, and minimize exposure to plastics.
Causes for Rejection
Wet/frozen tissue, cytology smears, nonformalin fixed tissue, nonparaffin embedded tissue, noncharged slides, ProbeOn slides, Snowcoat slides.
