Use
This immunohistochemistry test evaluates BRCA1 associated protein‑1 (BAP1) expression in tissue specimens. Loss of nuclear BAP1 staining can indicate inactivation of this tumor suppressor and is useful in differentiating malignant processes such as mesothelioma and select carcinomas, although interpretation requires correlation with clinical and histologic context.
Special Instructions
This test is a laboratory developed test (LDT) performed as a stain‑and‑return technical service only. Ordering clients should refer to the ARUP Immunohistochemistry Stain Offerings brochure. Requisition forms and interface maps are needed for build; component codes cannot be used for order entry.
Limitations
Only technical staining is performed; no interpretive pathology is included. Loss of staining cannot differentiate germline from somatic mutations. The test relies on representative tissue; non‑representative or depleted specimens are unacceptable. Optimal staining requires appropriate formalin fixation and slide handling (eg, avoid oven baking), and specimen quality (eg, slide age, environmental exposure) may impact results.

Formalin‑fixed paraffin embedded block or unstained slides (min: 2 slides)
Collection Instructions
Formalin fix (10 percent neutral buffered formalin) and paraffin embed specimen (cells must be prepared into a cellblock). Transport tissue block or 5 unstained (3‑ to 5‑micron thick sections), positively charged slides in a tissue transport kit (ARUP supply #47808). If sending precut slides, do not oven bake.
Storage Instructions
Protect paraffin block and/or slides from excessive heat; ship in cooled container during summer months.
