Use
This test is used for the assessment of pruritic, urticarial, blistering, and/or erosive inflammatory conditions, including immune-mediated cutaneous diseases. Direct Immunofluorescence is particularly useful in diagnosing immunobullous diseases, epithelial antibody-associated disorders, and other inflammatory skin conditions by detecting specific immunoglobulin deposits in biopsy specimens.
Special Instructions
Optimal specimen location and type, along with complementary serum testing and histopathological examination, are crucial for accurate diagnosis. Ensure that tissues are transported in the appropriate medium (Michel's medium or Zeus tissue fixative) and labeled properly if not using ARUP-supplied vials. Avoid formalin-fixed or frozen tissue in Michel's medium, as these conditions are unacceptable.
Limitations
Direct Immunofluorescence may not distinguish between certain pemphigoid variants or differentiate specific immunological markers in isolation; additional serum testing or alternative fixation methods may be required for comprehensive analysis. The fluorescence fades over time, limiting long-term specimen visualization. Negative control sections and technical adequacy checks are essential for ensuring valid results.
