FUS (16p11) Gene Rearrangement by FISH
Also known as: FUS FISH
Use
The FUS (16p11) Gene Rearrangement by FISH test is used to detect translocations involving the FUS gene. It is particularly useful for identifying low-grade fibromyxoid sarcoma (LGFMS) and distinguishing myxoid liposarcoma/round cell liposarcoma from other soft tissue tumors. A positive FUS rearrangement supports the diagnosis of LGFMS. The test is based on the performance during validation and is expected to detect 100 percent of FUS rearrangements in patients with FUS rearranged tumors, except for rare instances of cryptic rearrangements.
Special Instructions
Use this test in conjunction with histologic evaluation. Ensure specimen involves formalin fixation and paraffin embedding of tumor tissue. If multiple specimens are sent, a pathologist block selection fee may apply.
Limitations
This test is designed to detect rearrangements in the FUS gene but does not identify specific partner genes. It requires correlation with clinical and histopathologic findings due to variability in presentation. The assay does not detect all possible rearrangements, and false negatives can occur in cases of cryptic rearrangements. Not FDA-cleared or approved, though it performed in a CLIA-certified laboratory.
