SDHB with Interpretation by Immunohistochemistry

Use
The test screens for potential succinate dehydrogenase (SDH) germline mutations in patients with specific tumor types. Abnormal immunohistochemical staining for succinate dehydrogenase B (SDHB) correlates with the presence of a germline mutation in the genes of the succinate dehydrogenase complex, namely SDHB, SDHC, and SDHD, although it is not definitive. Genetic counseling is advised for further investigation when abnormal results are found. More comprehensive genetic testing is suggested to confirm findings and assess other genes associated with abnormal staining, such as SDHAF2 and SDHA.
Special Instructions
Submit electronic requests for immunohistochemistry. If multiple specimens are sent, specify if ARUP's pathologist should choose the most appropriate specimen or submit individual orders for each sample. A Pathologist Block Selection Fee may apply. Include the surgical pathology report with the specimen and indicate the tissue site on the requisition.
Limitations
This test has been developed and validated by ARUP Laboratories but it has not been cleared or approved by the FDA. Testing results must be correlated with clinical findings and patient history for accurate interpretation. The test is performed in a CLIA certified laboratory and is intended for clinical purposes. Mutations in genes other than those specifically targeted (SDHB, SDHC, SDHD) may also affect the results, indicating the need for comprehensive genetic assessment.

Collection Instructions
Formalin fix (10 percent neutral buffered formalin) and paraffin embed specimen. Protect from excessive heat and use ARUP Supply #47808 for transportation.

Room temperature or refrigerated. Ship in cooled container during summer months.
Causes for Rejection
Paraffin block with no tumor tissue remaining, specimens fixed in non-formalin fixatives, and decalcified specimens.
