Use
This test is essential for diagnosing mitochondrial diseases resulted from sequence variants or significant deletions/duplications in mitochondrial DNA (mtDNA). It is indicated for individuals suspected of mitochondrial disorders, and the presence of specific mutations can impact clinical management and familial risk assessments. The test provides a comprehensive view of mitochondrial variants, allowing for the detection of heteroplasmy levels which significantly affect phenotypic expressions. The maternal inheritance pattern of mtDNA also plays a crucial role in the interpretation of results, factoring in potential de novo changes that might occur in somatic tissues.
Special Instructions
The test employs next-generation sequencing (NGS) to analyze the entire mitochondrial genome. Recommendations include consulting with our laboratory if there are concerns about heteroplasmy or specific deletion/duplication analysis, as test 3056 might be more appropriate under such circumstances.
Limitations
While this analysis covers the entire mitochondrial genome, the accuracy of heteroplasmy measurements diminishes for large deletions due to preferential amplification biases. The test may identify variants present in asymptomatic individuals, which may not have clear clinical correlates. Additionally, small insertions and deletions under 10 base pairs are subject to estimation inaccuracies. Some rare genetic variants identified might not have definitive clinical relevance due to sparse representation in the existing databases, emphasizing the necessity for careful interpretation of findings within their clinical context.
