Tay-Sachs Disease Carrier Testing (Serum)
Use
Tay-Sachs disease carrier testing is crucial for screening individuals, particularly those of Ashkenazi Jewish, French Canadian, or Cajun descent, who may unknowingly carry the HEXA mutations leading to Tay-Sachs disease. This test analyzes hexosaminidase A levels in serum to identify carriers of Tay-Sachs and Sandhoff diseases. Affected individuals exhibit severe neurological deterioration, typically starting in infancy, making early detection vital for family planning. The presence of low HexA activity indicates carrier status, but molecular testing is recommended to clarify results as some variants may not be detectable through enzyme activity alone. Negative screening results indicate a reduced risk but do not entirely eliminate the possibility of having an affected child with a carrier partner.
Special Instructions
This test is suitable for individuals planning a pregnancy, those with a family history of Tay-Sachs disease, and individuals of Ashkenazi Jewish, French Canadian, or Cajun descent. For pregnant individuals, it is advisable to use test code 4620, which tests white blood cells instead of serum. Carrier testing can serve as a valuable tool for couples considering having children, providing insights into the likelihood of having an affected child based on carrier status.
Limitations
A small number of false positive results may occur due to pseudodeficiency alleles, presenting HEXA activity in carrier ranges. Genetic testing can help clarify uncertainty, as certain variants may evade detection through enzyme activity tests. In addition, the test cannot be used for pregnant patients. Individuals with little to no enzyme activity may still carry certain genetic mutations undetected by this method, making genetic counseling essential for thorough risk assessment and family planning.
