Use
Karyotyping, physical localization of copy number changes, and mapping of breakpoints involved in translocations.
Special Instructions
Include a tentative diagnosis or treatment plan on the test request form to facilitate proper culturing and result interpretation. If the Plasma-enriched Multiple Myeloma (MM) Profile, FISH [510325] is ordered concurrently and the sample is insufficient for both tests, the FISH test will take priority.
Limitations
The use of lithium heparin or EDTA anticoagulants can inhibit cell culture growth, which would affect the test's outcomes. Only samples with more than 4% blasts are suitable, except in cases of CLL, hairy cell leukemia, and small-cell lymphocytic leukemia where β-mitogen stimulation can be applied. Nonnormative specimens, gross contamination, and clotted or fixed samples are causes for rejection. Also, this analysis may not detect clonal molecular alterations below the resolution of light microscopy; additional molecular or FISH testing may be necessary.

Storage Instructions

Causes for Rejection
Hemolysis; gross contamination; non-neoplastic cells; clotted specimen; frozen specimen; fixed specimen; improper or too much anticoagulant.
