Use
This test is useful for the rapid detection of Coccidioides DNA, providing aid in the diagnosis of coccidioidomycosis using paraffin-embedded tissue specimens. Coccidioidomycosis is caused by the dimorphic fungi, Coccidioides immitis and Coccidioides posadasii, which are endemic to the southwestern United States and other regions. The test facilitates quick diagnosis compared to culture methods, which can take weeks and pose significant safety hazards to laboratory personnel due to the propagation of the organism.
Special Instructions
Specimen source is required. Body tissue specimens should be submitted preferably as formalin-fixed, paraffin-embedded tissue blocks. Each cut section must be placed in a separate sterile container for submission. The specimen must be ambient (preferred) or refrigerated during transit to the laboratory.
Limitations
The test detects Coccidioides nucleic acid, but does not distinguish between viable organisms or nucleic acid from past infections. Results should be correlated with patient history and symptoms. A negative result does not rule out an active infection because organism levels may be below the detection limit, and fixation can reduce sensitivity. The test cannot differentiate between Coccidioides immitis and Coccidioides posadasii.

Collection Instructions
Submit a formalin-fixed, paraffin-embedded tissue block to be cut and returned. Perform microtomy and prepare five separate 10-micron sections, each in a separate sterile container.
