Use
This test is used for determining overexpression of HER2 protein in gastric and esophageal adenocarcinoma using formalin-fixed, paraffin-embedded tissue sections. HER2 overexpression is associated with certain prognostic outcomes, including shorter disease-free survival and overall survival, in not only gastric and gastroesophageal junction cancers but also other cancers such as breast, ovarian, and endometrial cancers.
Special Instructions
For diagnostic purposes, order PATHC/Pathology Consultation and request the stains. For specimens without metastatic adenocarcinoma from the stomach or esophagus, different tests may be required (e.g., H2BR HER2 Amplification, Miscellaneous Tumor, FISH, Tissue). For breast cancer specimens, order a different test such as HERBA or HERBN. Include a pathology report stating the final diagnosis or a preliminary diagnosis referencing gastric or esophageal adenocarcinoma.
Limitations
This test does not have a reflex to FISH testing. Performance depends on proper fixation of the 10% neutral-buffered formalin-fixed paraffin-embedded tissue. It is recommended to fix surgical specimens between 18-24 hours and biopsy specimens between 6-8 hours.
