Use
This test aids in the diagnosis of giant cell tumor of bone by detecting the presence of the Histone 3.3 G34W mutation. The mutation is expressed in the nuclei of the mononuclear cell population of H3F3A altered giant cell tumor of bone, providing a valuable marker for both benign and malignant forms of this condition.
Special Instructions
This test only covers the technical component of the staining process and does not provide pathologist interpretation. It should not be ordered with a Pathology Consultation (PATHC), as the stains will be canceled if combined with PATHC. Specimen should be shipped with appropriate labels and must not be ordered as part of a consultation without discretion of the reviewing pathologist.
Limitations
This test does not include pathologist interpretation. Only technical performance is provided. The immunoreactivity of paraffin sections may be affected by the age of the sections, with best practices suggesting cutting within 6 weeks. Additionally, the chemistry of glass slides can influence staining, especially if exposed to heat, humidity, or improper storage conditions.

Formalin-fixed, paraffin-embedded tissue block or two unstained, positively charged glass slides (25- x 75- x 1-mm) per test ordered; sections 4-microns thick.
Causes for Rejection
Wet/frozen tissue; cytology smears; nonformalin fixed tissue; nonparaffin embedded tissue; noncharged slides; ProbeOn or Snowcoat slides.
