Lung Cancer, RET (10q11) Rearrangement, FISH, Tissue
Use
Identifying RET gene rearrangements in patients with late-stage, lung adenocarcinomas that are negative for epidermal growth factor receptor mutations and anaplastic lymphoma kinase rearrangements. It also supports the diagnosis of certain salivary gland, cutaneous, and other tumors when used in conjunction with an anatomic pathology consultation. Tumors with RET fusions may be sensitive to directed inhibitor therapy, suggesting potential response to targeted treatments.
Special Instructions
A pathology report is required for testing. Acceptable pathology reports include working drafts, preliminary pathology, or surgical pathology reports. The tissue block submitted should preferably be formalin-fixed, though attempts will be made with other fixation methods, albeit with less favorable outcomes.
Limitations
This test is not approved by the U.S. Food and Drug Administration and is used as an adjunct to existing clinical and pathologic information. Fluorescence in situ hybridization (FISH) does not rule out other chromosomal abnormalities. Fixatives other than formalin may not be successful for FISH assays. Decalcified paraffin-embedded tissues have a lower success rate for FISH analysis. Insufficient cell counts or inadequate tissue may result in no analysis charges but may also impair result interpretation.
