Phosphoglycerate Kinase Enzyme Activity, Blood
Use
The Phosphoglycerate Kinase Enzyme Activity test is useful for the evaluation of individuals with Coombs-negative nonspherocytic hemolytic anemia, especially if there is a pattern of X-linked inheritance. It is also important for patients with myopathic or neurologic symptoms as it helps in diagnosing phosphoglycerate kinase deficiency, an X-linked disorder. This enzyme plays a critical role in glycolysis by converting 1,3-diphosphoglycerate to 3-phosphoglyceric acid, a step that generates adenosine triphosphate (ATP).
Special Instructions
If electronic ordering is not available, a Benign Hematology Test Request Form (T755) must be completed and sent with the specimen. Ensure the specimen is collected in the recommended container and volume to avoid rejections. Specimens must not be aliquoted.
Limitations
Phosphoglycerate kinase deficiency may not always correlate with the enzyme activity levels. Female heterozygous individuals may show mild symptoms. Recent transfusions or reticulocytosis may affect the enzyme activity measurements. It's important to compare the activities of other red blood cell enzymes to avoid diagnostic errors.
