Use
This test is used to determine male fertility status and select the most cost-effective therapy for treating male-factor infertility. It quantifies the number of germinal and white blood cells per milliliter of semen, which is crucial for diagnosing male-factor infertility. Abnormalities found in sperm morphology could affect sperm transport and fertilization abilities, leading to the recommendation of specific treatments such as in vitro fertilization (IVF) or intracytoplasmic sperm injection.
Special Instructions
This test should not be used to check patient's sterility following a vasectomy. Patients should have 2 to 7 days of sexual abstinence at the time of semen collection for accurate results. Specimens must arrive within 24 hours of collection, and they should be sent from Monday through Thursday.
Limitations
Results might be affected by improper specimen transportation. The test does not check for sterility after vasectomy or fructose testing due to the use of dilution media. Multiple analyses might be required to confirm fertility status due to variability in semen parameters.

Collection Instructions
Allow dilution medium to warm to room temperature for 45 to 60 minutes. Allow semen to liquefy at room temperature for up to 30 minutes. Use sterile volumetric pipet or tube for volume measurement. Pour liquefied semen into 50-mL dilution medium container within 60 minutes of semen collection time, cap tightly, but do not overtighten, and gently mix.
Patient Preparation
Patient should have 2 to 7 days of sexual abstinence at the time of semen collection for accurate results.
Storage Instructions
Proper temperature maintenance of specimen throughout processing and shipping is critical. All materials the specimen is exposed to should be at room temperature (20–28 degrees C).
Causes for Rejection
All specimens will be evaluated at Mayo Clinic Laboratories for test suitability.
